The fourfold assembly originally referred to those who gathered to hear the Buddha teach the Dharma: Bhikshus, Bhikshunis, upāsakas, and upāsikās, in other words, monks and nuns, and laymen and laywomen. It now also refers in general to those four categories of Buddhists.
